PEAC Biomedical Resource


 

PEAC biomedical resource is a unique and comprehensive collection of samples from patients with early Arthritis together with high-density data including genomic and transcriptomic analysis, biologic tissue characterisation, state-of-the-art 3/4D ultrasound imaging coupled with detailed clinical phenotyping. This resource is available to researchers through application to the PEAC Steering committee.

All PEAC samples are collected with full approval of the relevant research ethics committees and full consent of the tissue donor. All PEAC samples are coded.


Synovial Tissue & Synovial fluid

Synovial tissue and synovial fluid is obtained through biopsies. Synovial cellular phenotypes (T-B-Macrophage-FDC etc.) and lymphoid organisation are determined by immunohistochemistry (IHC) and digital image analysis. Biopsies are classified according to the aggregational tendency and the acquisition of the typical features of secondary lymphoid organs.


Serum & Synovial fluid cytokines and chemokines analysis.

Cytokines, chemokines and growth factors (24 moieties measured per 50μl sample) are measured in Synovial fluid and serum samples using a multiplex detection method (Luminex).


Gene expression profiling

Transcriptional analysis of synovial tissue samples is examined via Micro-array based gene expression profiling using the Illumina micro-array platform.


Proteomics and Metabonomics

Bio samples are collected and stored in preservatives for Proteomics and Metabonomics studies.


Genotyping

  • DNA: DNA is extracted and quality controlled prior to archiving at the PEAC DNA repository.
  • RNA: RNA is extracted and quality controlled prior to archiving at the PEAC RNA repository. Samples are further processed for cDNA.
